What is a peptide?

A peptide is a short chain of amino acid residues linked by covalent amide (peptide) bonds, formed via condensation reactions between the α-carboxyl group of one amino acid and the α-amino group of the next. By convention, a peptide typically contains between 2 and approximately 50 amino acid residues; longer chains are generally classified as proteins.

The linear sequence of residues, written from the N-terminus (free α-amino group) to the C-terminus (free α-carboxyl group), defines the peptide’s primary structure. This sequence determines the molecule’s physicochemical properties — molecular weight, isoelectric point (pI), hydrophobicity, and net charge at a given pH — as well as its three-dimensional conformation and biological activity in receptor binding, enzymatic, and signaling pathways.

Peptides are synthesized in the laboratory primarily by solid-phase peptide synthesis (SPPS), wherein residues are coupled stepwise onto an insoluble resin support, followed by cleavage and purification. Purity and identity are then characterized analytically — typically by reversed-phase high-performance liquid chromatography (RP-HPLC) for purity, and mass spectrometry (MS) for molecular mass and sequence confirmation.
